Do you mean that you want to run the LL test, which tests the number for primality, or that you want to do all the work for one number from start to end yourself?
To run LL tests, set Prime95 to request those instead of TF in Test > Worker Windows.
To do all the work for a number, go to http://www.mersenne.org/manual_assignment/, log in, choose "Smallest available first-time tests", and specify which exponent(s) you want to do using the exponent range boxes (if you leave those blank, it will give you the smallest LL test available). If you want more than one, change the "Number of CPUs..." field to how many you want. Then if the number is available, it will give you a line to put in your worktodo.txt file. Copy/paste that to the end of the worktodo.txt file for the worker number that you want to run it, then restart Prime95 to make it see the change.

Normal GIMPS assignments have the separate parts of testing a single number (trial factoring, P-1 to find small factors, the LL test to check primality, and a second LL test to double check that the first one was right) assigned and done separately.
