GPU setup

Is there a FAQ anywhere on the setup of using a GPU? I have tried searches and looking in several threads, but I cannot find a good explanation on how to get running on this.
I am running a 32 bit Windows Vista machine with a Core 2 Duo and a GeForce GTS 450.
